Cer sequence from plasmid ColE1
This part consist of the cer sequence from the plasmid ColE1. It is a component from the cer-xer system that converts dimers by recombination back to monomers. This stabilizes the transmission of the plasmid that contains it even when no antibiotic is added to the growth media. These properties of the cer sequence have been described multiple times in the literature.
To test the plasmid-stabilizing properties of our new part, we had it cloned into a pSB1C3 vector with a red colour device. Positive clones were identified by colony PCR and then cultivated overnight at 37oC using LB-media without antibiotics. On the next morning smmall aliquots of the overnight cultures were plated on LB-plates with chloramphenicol. As one can see on the picture, the cer containing version of the pSB1C3 vector shows improved stability.
This part has also an inverted version - BBa_K2847001. We submitted it since the inverted version can be more efficient in stabilizing different plasmid constructs.
